How to calculate the risk ratio(Enrichment) of mutations on interaction interfaces

Hi there, I’m doing a study about the impact of mutations on the interaction interface of proteins, and I need to calculate the risk ratio between the disease group and the control group. I found a published paper that might have a solution for my question, however, I can’t figure out how did they get their results.

I tried to find a way to calculate the enrichment of different groups with their data. However, I got the wrong enrichment every time, I have no clue how to calculate the enrichment. Any idea? Any suggestion will be appreciated! Thank you!
